Genotoxic Potentials of Natural Products Detected by a Short-Term Test Using Diploid Strains of Aspergillus nidulans

چکیده
Main aim is to evaluate the genotoxic potentials of three classes of natural products, a sesquiterpene lactone (cynaropicrin), a saponin (stigmast-7en-3 -ol-glucopyranoside) and a steroid (stigmasterol), at low doses, using the Homozygotization Index test (HI). This short-term test, deals with the determination of alterations in mitotic crossing-over frequencies eventually involving heterozygous auxotrophic genes in diploid strains of Aspergillus nidulans. In this way, treatments of UT448//UT184 and Dp II-I//UT184 diploid strains with the natural products showed that saponin and lactone significantly increased the homozygotization index (HI) of both strains, especially for the diploid Dp II-I//UT184. No effect was observed with the steroid. On this account it presents remarkable potentials to detect the genotoxic/carcinogenic effects of bioactive natural products. Furthermore, this is the first time the low doses effects of bioactive natural products were detected using A. nidulans.

Genotoxic evaluation of sodium nitroprusside in Aspergillus nidulans
The exogenous nitric oxide donor, sodium nitroprusside, evaluated the recombinogenic potential of nitric oxide. Drug inhibited mycelial growth and conidiation in A757 Aspergillus nidulans master strain. Two heterozygous diploid strains, one wild (uvsH//uvsH) and the other defective to DNA repair (uvsH//uvsH) were used for recombinagenesis tests. Sodium nitroprusside recombinogenic effect was ev...
متن کاملAspergillus nidulans as a biological system to detect the genotoxic effects of mercury fumes on eukaryotes.
Mercury (Hg) pollution is one of the most serious environmental problems. Due to public concern prompted by the symptoms displayed by people who consumed contaminated fish in Minamata, Japan in 1956, Hg pollution has since been kept under constant surveillance. However, despite considerable accumulation of knowledge on the noxious effects of ingested or inhaled Hg, especially for humans, there ...
متن کاملComparing artificial and natural selection in rate of adaptation to genetic stress in Aspergillus nidulans.
In an experimental study of adaptation to negative pleiotropic effects of a major fungicide resistance mutation in the filamentous fungus Aspergillus nidulans we have investigated the relative effectiveness of artificial selection vs. natural selection on the rate of compensatory evolution. Using mycelial growth rate as a fitness measure, artificial selection involved the weekly transfer of the...
متن کاملBenznidazole-induced genotoxicity in diploid cells of Aspergillus nidulans.
Genotoxic effects of benznidazole were studied by the induction of homozygosis of genes previously present in heterozygous. UT448//A757 diploid strain was used in the benznidazole's recombinagenesis test. Although toxic effects on growth of colonies were not observed, 75 and 100 microM benznidazole induced an increasing of mitotic recombination events in diploid strain. Results were related to ...
متن کاملInduction of mitotic crossing-over in diploid strains of Aspergillus nidulans using low-dose X-rays.
As a contribution towards detecting the genetic effects of low doses of genotoxic physical agents, this paper deals with the consequences of low-dose X-rays in the Aspergillus nidulans genome. The irradiation doses studied were those commonly used in dental clinics (1-5 cGy). Even very low doses promoted increased mitotic crossing-over frequencies in diploid strains heterozygous for several gen...
